Abstract

ABSTRAK: Bahan tambahan yang digunakan dalam formulasi sediaan farmasi seringkali mempengaruhi sifat kimia fisika bahan aktif. Propilen glikol adalah bahan yang banyak digunakan dalam formulasi sediaan semipadat, sediaan cair dan trsasdermal sebagai kosoIven, penambah viskositas dan plasiisizer. Penelitian ini mengkaji pengaruh propilen glikol terhadap kelarutan semu teofilin dan kofein dalam dapar fosfat pH 7,4. Penaltian dilakukan dengan sejumlah teofilin dan kofein dilarutkan dalam medium dapar fosfat pH 7,4 dengan kandungan propilen glikol sebesar 0, 10, 20, dan 30% v/v. Kelarutan teofilin dan kofein ditentukan secara spektrofotometri UV pada panjang gelombang 271 rim dan 274 nm. Hasil penelitian menunjukkan keberadaan propilen glikol dalam medium mailing- katkan kelarutan molar semu teofilin dan kofein. Peningkatan kelarutan karena pengaruh propilen glikol pada kofein lebih besar daripada teofilin. Hal ini teijadi karena energi interaksi solut solven kofein lebih rendah dan lebih banyak jurnlah molekul kofein yang tidak terionkan berada dalam larutan daripada teofilin. Kata kunci : teofilin, kofein, kelarutan semu, propilen glikol ABSTRACT: Additional substances used in the formulation of pharmaceutical dosage form often influence physicochemical parameters of drug. Propylene glycol is usually used in liquid, semisolid and transdermal formu lation as cosolvent, viscosity increasing agent, and plas- tizicer. The research was done to study the influence of propylene glycol on theophylline and caffeine apparent solubilities in phosphate buffer solution pH 7.4. In research, certain amounts of theophylline or caffeine were solubilized into phosphate buffer solution pH 7.4 containing propylene glycol at the concentrations of 0, 10, 20, 30 % v/v. The apparent solubility of theophylline and caffeine were measured by a UV spectrophotometric method, at the wave lengths of 271 nm (theophylline) and 274 run (caffeine). The results showed that propylene glycol increased theophylline and caffeine apparent solubilities. Caffeine apparent solubility was increased higher than that of theophylline, because of caffeine solute-solvent interactions energy was lower than that of theophylline, and there are more unionized caffeine in the solution than theophylline.
